    学位論文の研究テーマとなったアスコルビン酸(ビタミンC)が関与する様々な反応,アスコルビン酸の酸化分解経路の解明やアスコルビン酸が関与する生体内メイラード反応などをメインテーマとし,現在も研究活動を続けている。現在はこれらの研究に加えて,茨城県の食教育のあり方や高齢者に対する食教育の可能性など食教育関連の研究も手がけている。

  • Chemical Characteristics of the Proteinaceous Material in the Nests of Six Polistine Species (Hymenoptera; Vespidae)
    Takaaki Kuwahara; Sôichi Yamane; I-Hsin Sung Sirinee Poonchaisri; Yoko Nishikawa, Some chemical characteristics of oral secretions and larval silk, which contain proteinaceous substances, were analyzed in the nests of four Ropalidia species, R. fasciata, R. taiwana, R. rufocollaris and R. flavopicta, and two Polistes species, P. gigas and P. tenebricosus. Nitrogen contents in pure oral secretions ranged between 13-14%, and crude protein contents, calculated from the N contents, were 84-89%. The protein content in the larval silk was approximately 90%. The three major amino acids in the nest parts were Gly (23-48%), Ala (16-22%) and Ser (6-19%) in Ropalidia species, and Gly (20-32%), Ser (11-14%) and Pro (10-13%) in Polistes species. These items occupied about 60% of the residues. The three major amino acids in the larval silk were Ala (33-34%), Ser (19-21%) and Glx or Asx (9%). Ala and Ser comprised about half of the residues. In some species, the relative abundance of amino acids in the oral secretions varied depending on the parts of nest, especially in the proportion of Gly. Cluster analyses carried out on the amino acid compositions in the oral secretions generated two distinct groups: One consisted of the nest paper, which contained a small amount of secretion coating and the other consisted of secretion layers of the petiole and secretion membranes made at the bottom of Ropalidia cells. These results suggest a possibility that these wasps can produce oral secretions with different kinds and/or ratios of protein(s) according to their purposes, i.e., construction or maintenance., CALIFORNIA STATE UNIV

  • A study on systematic education to master the basics of cooking : Focusing on cutting skill               
    Sayaka HOSHI,Yoko NISHIKAWA, 家庭科では,近年の著しい授業時数の削減により,技能習得のための十分な実技教育ができない状況が生じている。調理における基礎技能習得は,主に小学校のうちに行われるのが望ましいが,この技能教育における教育時間と必要性のギャップを現行の小学校教育の中で埋めていくには,家庭科だけではなく他の授業時間の利用が有効と考えられる。その可能性を探るため,本研究では包丁技能習得教育を中心に,文献調査及び小学生を対象とした包丁技能テストを行い検討した。その結果,高学年の家庭科の授業を待たずとも,低学年のうちから教育は可能であり,成長段階に応じて内容を吟味すれば高い教育効果が得られると推察された。また,現在の家庭科教科書に掲載されている調理実習内容は,そのほとんどが調理科学的視点から編集されたものであり,技能習得に視点を置いた教科書の見直しが必要であると考えられた。

  • Vitamin C metabolomic mapping in the lens with 6-deoxy-6-fluoro-ascorbic acid and high-resolution 19F-NMR spectroscopy.

Metabolomics, or metabolic profiling, is an emerging discipline geared to providing information on a large number of metabolites, as a complement to genomics and proteomics. In the current study, a fluorine-labeled derivative of ascorbic acid (F-ASA), a major antioxidant- and UV-trapping molecule in the aqueous humor and the tens, was used to investigate the extent to which the lens accumulates potentially toxic degradation products of vitamin C.
    METHODS. Human lens epithelial cells (HLE-B3) and rat lenses were exposed to hyperglycemic or oxidative stress in vitro or in vivo and probed for accumulation of F-ASA, fluoro-dehydroascorbate (F-DHA), fluoro-2,3-diketogulonate (F-DKG), and their degradation products in protein-free extracts, by proton-decoupled 750-MHz F-19-nuclear magnetic resonance (NMR) spectroscopy.
    RESULTS. F-ASA and F-DHA were taken up into HLE B-3 cells by an Na+-dependent transporter. Their uptake was unexpectedly only slightly affected by hyperglycemia in vitro, unless glutathione was severely depleted. Glycemic stress catalyzed oxidation of F-ASA into a single novel F-compound at -212.4 ppm, whereas F-DHA and F-DKG were the major degradation products observed after GSH depletion. In contrast, F-ASA uptake was markedly suppressed in diabetic cataractous rat lenses, which accumulated both the F-DHA and the - 212.4-ppm compound. In an unexpected finding, the latter formed only from F-ASA and not F-DHA or F-DKG, suggesting a novel pathway of in vivo F-ASA degradation. Both the cells and the intact rat and human lenses were permeable to several advanced F-ASA and F-DHA degradation products, except F-DKG. The unknown compound at -212.4 ppm was the only F-ASA degradation product that spontaneously formed in rabbit aqueous humor upon incubation with F-ASA.
    CONCLUSIONS. These studies suggest the existence of a novel ascorbic-acid-degradation pathway in the lens and aqueous humor that is influenced by the nature of the oxidant stress. Under similar culture conditions, intact lenses are more prone to hyperglycemia-mediated oxidant stress than are lens epithelial cells, but both are permeable to various F-ASA degradation products, the structure and biological roles of which remain to be established., ASSOC RESEARCH VISION OPHTHALMOLOGY INC

Here we have probed ascorbic acid homeostasis and degradation in diabetes using 6-deoxy-6-fluoro ascorbic acid (F-ASA) and 750 MHz F-19-nuclear magnetic resonance (NMR) spectroscopy with proton decoupling In vitro, Cu2+-mediated degradation of F-ASA revealed the formation of 4 major stable degradation products at 24 hours. However, when normal or diabetics rats were injected with F-ASA intraperitoneally (IP) for 4 days, up to 20 fluorine-labeled compounds were observed in the urine. Their composition resembled, in part, metal catalyzed degradation of F-ASA and was not explained by spontaneous degradation in the urine. Diabetes led to a dramatic increase in urinary F-ASA loss and a relative decrease in most other urinary F-compounds. Diabetes tilted F-ASA homeostasis toward oxidation in liver [P < .01), kidney (P < .01), spleen (P < .01), and plasma (P < .01), but tended to decrease oxidation in brain, adrenal glands, and heart. Surprisingly, however, besides the major oxidation product fluoro-dehydroascorbic acid (F-DHA), no F-ASA advanced catabolites were detected in tissues at 5 mu mol/L sensitivity. These findings not only confirm the key role of the kidney in diabetes-mediated loss of ascorbic acid, but demonstrate that only selected tissues are prone to increased oxidation in diabetes. While the structure of most degradation products needs to be established, the method illustrates the power of high resolution F-19-NMR spectroscopy for the mapping of complex metabolomic pathways in disease states.   • Identification of 3,4-dihydroxy-2-oxo-butanal (L-threosone) as an intermediate compound in oxidative degradation of dehydro-L-ascorbic acid and 2,3-diketo-L-gulonic acid in a deuterium oxide phosphate buffer.
    Nishikawa Y; Toyoshima Y; Kurata T., Dehydro-L-ascorbic acid (DAA), an oxidation product of L-ascorbic acid (vitamin C), is unstable in the neutral and basic pH regions. When DAA was incubated in a phosphate buffer with deuterium oxide (pH 7.4), it was degraded to form the main degradation compound, which was identified as 3,4-dihydroxy-2-oxobutanal (L-threosone). This compound was also formed from diketo-L-gulonic acid (DKG) in a phosphate buffer with deuterium oxide. L-threosone had reducing activity, probably due to its enolization, and is likely to have been involved in the formation of the reducing activity that was observed in aqueous DAA and DKG solutions. As a reactive dicarbonyl compound, L-threosone might also take some role in the cross-linking of tissue proteins that are formed in vivo in the Maillard reaction., TAYLOR & FRANCIS LTD

  • 6-Deoxy-6-fluoro-L-ascorbic acid: crystal structure and oxidative degradation.

We have studied the feasibility of using 6-deoxy-6-fluoroascorbic acid (6) for identification of ascorbic acid degradation products by F-19 NMR spectroscopy. Crystals of compound 6 from nitromethane belonged to the space group P2(1) with a = 5.547(2), b = 6.769(3), c = 9.302(2) Angstrom, beta = 91.80(3)degrees and Z = 2. Atomic coordinates, bond lengths and angles, hydrogen coordinates, anisotropic and isotropic displacement parameters were similar if not identical with those of native ascorbic acid. Similarly, UV properties and oxidation kinetics by CuCl2 at different pH values were essentially identical with ascorbic acid. Using 750 MHz F-19 NMR spectroscopy, five to six new fluorinated products were detected after overnight oxidation of 6 with Cu2+, suggesting that 6 may be a powerful and sensitive tool for assessment of its catabolism in vivo.   • Chemical characteristics of dehydro-L-ascorbic acid.
    Kurata T; Nishikawa Y, Dehydro-L-ascorbic acid (DAA) exists mainly in its C2 hydrated bicyclic form (5) in an aqueous solution, and monocyclic DAA (3), which is the expected reaction product immediately after the oxidation of AA, has not been observed by NMR spectroscopy, The formation mechanism for 5 from 3 and the stability of 5 were examined by the semi-empirical molecular orbital method (MOPAC). It was indicated that the protonation reaction was the key step in the formation of 5, therefore, the formation of 5 Is thought to be more difficult under physiological conditions which mostly involve in the neutral or slightly alkaline state. Erythorbic acid (EA) is the epimer of AA and has chemical characteristics very similar to those of AA, It is demonstrated in the present study by H-1-NMR that dehydro-L-ascorbic acid (DAA) was reduced by EA under neutral conditions but not acidic, and that dehydroerythrobic acid (DEA) was also reduced by AA under the same conditions.. These reactions also occurred at a low concentration dose to the concentration of AA in such biological tissue as the liver. Furthermore, the interconversion of DAA and AA at neutral pH and low concentration was also confirmed by radioluminography.
