  • 〔Major achievements〕Search for Expression Marker Genes That Reflect the Physiological Conditions of Blossom End Enlargement Occurrence in Cucumber
    Rui Li; Runewa Atarashi; Agung Dian Kharisma; Nur Akbar Arofatullah; Yuki Tashiro; Junjira Satitmunnaithum; Sayuri Tanabata; Kenji Yamane; Tatsuo Sato, Last, Blossom end enlargement (BEE) is a postharvest deformation that may be related to the influx of photosynthetic assimilates before harvest. To elucidate the mechanism by which BEE occurs, expression marker genes that indicate the physiological condition of BEE-symptomatic fruit are necessary. First, we discovered that preharvest treatment with a synthetic cytokinin, N-(2-Chloro-4-pyridyl)-N’-phenylurea (CPPU), promoted fruit growth and suppressed BEE occurrence. This suggests that excessive assimilate influx is not a main cause of BEE occurrence. Subsequently, the expression levels of seven sugar-starvation marker genes, CsSEF1, AS, CsFDI1, CsPID, CsFUL1, CsETR1, and CsERF1B, were compared among symptomatic and asymptomatic fruits, combined with and without CPPU treatment. Only CsSEF1 showed a higher expression level in asymptomatic fruits than in symptomatic fruits, regardless of CPPU treatment. This was then tested using fruits stored via the modified-atmosphere packaging technique, which resulted in a lower occurrence of BEE, and the asymptomatic fruits showed a higher CsSEF1 expression level than symptomatic fruits, regardless of the packaging method. CsSEF1 codes a CCCH-type zinc finger protein, and an increase in the expression of CsSEF1 was correlated with a decrease in the fruit respiration rate.   • 〔Major achievements〕Cultivation conditions affect the occurrence of blossom-end enlargement in cucumber
    Junjira Satitmunnaithum; Hikaru Muroi; Rina Ito; Yuki Tashiro; Antonius Febri Hendratmo; Sayuri Tanabata; Tatsuo Sato, Last, Postharvest blossom-end enlargement (BEE) in summer cucumber has a serious economic effect in Japan. Previous studies suggested that BEE is influenced by cultivation conditions and temperature during transportation. In this study, the relationship between postharvest BEE and growth parameters in plants under various sink–source balances created by defoliation and fruit thinning was determined using a growth analysis technique. Cucumber (Cucumis sativus L. ‘Taibo I’) seedlings were transplanted into an outdoor field at Ibaraki University in 2019, and the harvested fruits were stored in plastic bags at 27°C. The occurrence of BEE, which was scored six days after harvest, was positively correlated with the field air temperature and cumulative duration of sunshine exposure from flowering to harvesting. Furthermore, high temperatures and extended sunshine exposure reduced the time from flowering to harvesting. However, excessive defoliation reduced BEE occurrence, most likely because of the increased time from flowering to harvesting, suggesting that early fruit maturation may be one of the causes of BEE. Therefore, principal component analysis was performed to understand the relationship between growth parameters and the occurrence of BEE in cucumber. The occurrence of BEE increased when the sink–source balance was inclined towards the source. Collectively, these findings indicate that high temperatures, extended exposure to solar radiation, and large leaf area with respect to the number of fruit set increase the occurrence of BEE, with pre-harvest fruit maturity probably related to its onset., The Japanese Society for Horticultural Science

  • 〔Major achievements〕Upregulation of heat shock transcription factors, Hsp70, and defense-related genes in heat shock-induced resistance against powdery mildew in cucumber
    AniWidiastuti; Nur Akbar Arofatullah; Agung Dian Kharisma; TatsuoSato, Last, This study investigated heat shock-induced resistance (HSIR) against cucumber powdery mildew and confirmed the initial mechanism of HSIR prior to systemic acquired resistance. Heat shock treatment reduced the disease index of powdery mildew when the inoculation period was 24 h or less. HSIR increased the gene expression of Hsp70 12 h after the treatment, while the gene expression levels of POX and LOX6 increased 24 h after the treatment. Heat shock element (HSE) motifs were found in the promoter area of these target genes. The gene expression levels of HsfA2, HsfB1, and HsfB4 increased 6 h after heat shock. Therefore, upon heat stress, some heat shock transcription factors were activated to bind to HSEs and upregulate the transcription of Hsp70 and defense-related genes., Elsevier

  • 〔Major achievements〕Case Study on the Use of the Leaf-Count Method for Drip Fertigation in Outdoor Cucumber Cultivation in Reconstructed Fields Devastated by a Tsunami
    Yuki Tashiro; Tatsuo Sato; Junjira Satitmunnaithum; Hatsumi Kinouchi; Jianlin Li; Sayuri Tanabata, Corresponding, Drip fertigation was tested in fields using a nitrogen fertilization method based on leaf increments, with the aim of increasing the cucumber yield in outdoor fields restored after the tsunami disaster in Rikuzentakata City, Iwate Prefecture, in 2011. The 2016 test site (Takata field) was restored as a paddy field, and there were problems with water retention and gravel contamination. The condition of the 2017 test site (Yonesaki field) was better than that of the 2016 site. The drip fertigation method increased cucumber yield by 93% and 27% in the Takata and Yonesaki fields, respectively, when compared to the yield from fields cultivated conventionally. Drip fertigation enables the constant supply of liquid fertilizer to the rhizosphere, and the easy application prevents the scarcity of fertilizer, especially at later stages of growth. In contrast, a real-time soil diagnosis, using the Dutch 1:2 soil–water extract method, was unsuccessful due to flooding, especially in the Takata field. As the purpose of this method is not to reduce the amount of nitrogen provided, but to increase the yield, and because it is difficult to precisely control the application of fertilizer due to precipitation, we suggest that the real-time soil diagnosis and feedback should be omitted to further simplify fertilizer application., MDPI

  • Effects of organic fertilizer on bok choy growth and quality in hydroponic cultures

When OF and CF were applied with the same amount of total nitrogen in summer cultivation, there was no significant difference between yields; however, the growth rate in OF was slower than in CF. When OF was applied with twice the amount of nitrogen in CF (OF2), bok choy growth and yield were significantly inhibited in summer cultivation, likely owing to dissolved oxygen deficiency and different rates of nitrification and nitrogen absorbance by the plant root. Although the contents of potassium, calcium, and magnesium in bok choy showed no difference among the three treatments in both cultivation seasons, the carbon/nitrogen ratio tended to be higher in OF and OF2 than in CF. Lower nitric acid and higher ascorbic acid content was found in OF and OF2 than in CF. Overall, our results suggest that a comparable yield is expected by using the same nitrogen amount with a conventional recipe of chemical fertilization in autumn cultivation.   • Heat Shock-Induced Resistance against Pseudomonas syringae pv. tomato (Okabe) Young et al. via Heat Shock Transcription Factors in Tomato

Leaf disease symptoms were significantly reduced at 12 and 24 h after HST, consistent with upregulation of pathogenesis-related (PR) genes PR1a2 and PR1b1 peaking at 24 h after treatment. These genes were upregulated at the treatment application site, but not in untreated leaves. In contrast to HST, inoculation of the first leaf induced systemic upregulation of acidic PR genes in uninoculated second leaves. Furthermore, heat shock element motifs were found in upstream regions of PR1a2, PR1b1, Chitinase 3, Chitinase 9, Glucanase A, and Glucanase B genes. Upregulation of HsfA2 and HsfB1 peaked at 6 h after HST, 6 h earlier than salicylic acid accumulation. Foliar spray of heat shock protein 90 (Hsp90) inhibitor geldanamycin (GDA) induced PR gene expression comparable to that after HST. PR gene expression and defense response against Pseudomonas syringae pv. tomato (Pst) decreased when combining HST with Hsfs inhibitor KRIBB11. The Hsfs and PR gene expression induced by heat or GDA, together with the suppression of heat shock-induced resistance (HSIR) against Pst by KRIBB11, suggested a direct contribution of Hsfs to HSIR regulation in tomato., MDPI

    Mioko Yoshino; Tatsuo Sato; Ani Widiastuti; Tasuku Kato; Eiichi Inoue; Masahide Iwata; Kenji Fukushi; Koji Iwashita; Fumio Watanabe, Corresponding, メタン発酵消化液(消化液)は肥料として有用な窒素成分を含むが,不溶性成分も含み粘稠性が高いため,簡便,均一に施用することが難しく,施設園芸分野での利用は進んでいない. そこで,スクリーンフィルターを改造して不溶性成分を除去する濾過装置と,水圧を利用して灌水に消化液を混入する施肥装置を試作し, 雨よけキュウリの灌水同時施肥栽培での適用を試みた. 期間ごとの消化液の施用量は,葉数に基づく窒素施肥指標により調節した. 対照として,尿素施用区を設けた. その結果,試作装置により消化液の物性の問題は改善され,作期を通して詰まりの問題もなく,灌水同時施肥栽培が可能であった. 葉数の推移,施肥量,収量についても,処理間で明らかな差は認められなかった. 施用した消化液は速やかに分解,硝化され,肥料としてキュウリに吸収されたものと考えられた. これらのことから,施用を適切に行うことができれば,本消化液は尿素と同等の肥効を持つ窒素肥料として利用可能であることが示唆された., 園芸学会

  • Improvement of Heat Shock Treatment and Trial Development of Hot Water Sprayer for Inducing Disease Resistance in Cucumber (Cucumis sativus L.)
    Mioko Yoshino; Ani Widiastuti; Zhou Songying; Hiromitsu Odani; Morifumi Hasegawa; Tatsuo Sato, Corresponding, 病害抵抗性誘導を目的とする熱ショック処理の連続・一斉処理が可能な方法の開発を目指して本研究を行った. 材料にキュウリ幼苗を用い,より短時間の処理条件を明らかにするとともに,温湯散布装置を試作してその効果を検証した. 温湯浸漬処理による灰色かび病接種試験の結果,50℃20秒の処理は,これまで最適条件とされてきた40℃120秒の処理と同等の抵抗性誘導効果を示した. そこで,LPガス給湯器を主構成とし,散布ノズルを保温カバーで覆った温湯散布装置を試作し,キュウリ幼苗に葉温が最高50℃となるように処理を施した. その結果,本装置を用いた処理により, 全身獲得抵抗性のシグナルを伝達する葉中サリチル酸含量は増加し,病害抵抗性関連遺伝子であるぺルオキシダ一ゼ遺伝子の発現レベルが上昇した. 灰色かび病接種試験では,抵抗性誘導剤として知られるBITと同程度の抵抗性を示した. 以上の結果から,これまでの報告より高温・短時間である50℃20秒の熱ショックにより,キュウリ幼苗に灰色かび病抵抗性を誘導でき,また温湯散布処理による熱ショックにおいても効果があることが確認されたことから,連続一斉処理技術の開発が可能であると考えられた., 園芸学会

    Mioko Yoshino; Ani Widiastuti; orifumi Hasegawa; Tatsuo Sato, Corresponding, The optimum conditions and mechanisms of inducing disease resistance by heat shock were investigated against gray mold (Botrytis cinerea) in cucumber (Cucumis sativus L.). The growth of inoculated mycelium was controlled most effectively by treatment at 40°C, for 2 minutes. Salicylic acid accumulated in the treated leaves under this condition and two peaks were obtained 24 and 96 hours after treatment. A pathogenesis-related protein, peroxidase gene (POX; M91372), was also expressed in the treated leaves and showed two peaks at 24 and 96 hours, respectively after treatment. Partial heat shock treatment was conducted by dipping only the first leaf of two leaves seedlings. Resistance and POX expression were induced not only in the treated leaves but also in the untreated second leaves. In conclusion, heat shock by means of hot water dipping could induce resistance against gray mold in cucumber. Systemic acquired resistance may have been obtained by some possible pathway.

  • The Effects of Liquid-coating Mulch Spray on Growth, Yield and Undersoil Temperature,in Komatsuna Greens (Brassica rapa L.)

Four treatments (1, 0.5, 0.25 L·m−2) of spray amount were tested in combination with seeding date (Sep. 21st, Oct. 5th, Oct. 20th in an open field and the Jan. 22nd in a plastic film house). As a result, we found this liquid-coating mulch increased yields of Komatsuna greens, unrelated to the spray amount. Generally, the daily highest soil temperature (−5 cm) under the mulching exceeded the control in every treatment however the daily lowest temperature were less than control except in the case of seeding at Sep. 21st. These phenomena were remarkable in the early stage of growing and the differences in temperature between the control and mulch treatments were reduced during the growth. There was no significant difference in the hourly-integrated temperature during the first 10 days between the 4 treatments on same seeding date. These findings suggest that the expansion of the daily soil temperature range contributed the increased yields of Komatsuna greens.

  • The estimation of absorbed nitrogen using increased leaf number in drip- fertigated spring cucumber
    Tatsuo Sato; Kyoko Matsuura; Jiro Narimatsu; Yutaka Yoneyama, Lead, Destructive analysis was used to estimate the half-monthly nitrogen content and related growth parameters of drip-fertigated spring cucumber.
    Three fertilizing treatments were compared. The amount of nitrogen applied half-monthly varied independently, but the total amount was same. There was little difference in the growth and yield of cucumber among nitrogen application patterns, which suggests that soil has a buffering capacity on cucumber yield even if the amount of nitrogen fertilizer changed. Destructive analysis including three all treatments found a significant correlation between nitrogen content of cucumber plant and the number of increased leaves (absorbed nitrogen (g/m2/14 days)=increased leaves (no./m2/14 days)×0.0441+2.189). This relationship will lead to reduce nitrogen application through counting of leaves twice a month., Japanese Society of Farm Work Research

  • 心止まり性トマト品種‘なつのこま’の加熱調理用トマトとしての栽培特性
    佐藤達雄; 石井孝典; 久保深雪; 成松次郎, Lead, A method of cultivating the tomato variety "Natsunokoma" on a small scale, in a suburban field, was tested from the viewpoint of longer period in harvest and saving labor. Early sowing and planting immediately after the last frost were required for a high yield. Multiple sowing will extend the harvest over several months, as the harvest period of each plant is short. The recommended planting spacing is 160×40cm (156 plants/a). The harvest period was not prolonged if the planting density was reduced. Harvesting once a week, and harvesting using hand-vibration, saved labor.
    These results suggested the following policies were useful for producing cooking use tomato: 'Natsunokoma'. (1) Multiple sowing was available from February until April. Planting and growth season should avoid the last frost and rainy season, respectively. (2) The recommended planting spacing is 160×40cm. Any training may be unnecessary. (3) Only red fruit should be harvested repeatedly by hand when the yield was little. A single harvest method by hand-shaking should be done in obedience to the marketing, labor intensity and the ripening., Japanese Society of Farm Work Research

Fruit yield for 5 cultivars of summer cucumber was increased by setting the ventilation temperature at 45℃ from 4:30 until 10:30. Diseases and insects were effectively suppressed by high temperature. Non-ventilation should be done during the lunch break during 11:30-13:20 in consideration of the worker's comfort. Most diseases and insects (except mites) could be controlled. However, with daily high-temperature treatment the number of dead or malformed fruits increased. Until the beginning of harvest, frequent non-ventilation treatment may exterminate diseases and insects and increase the number of fruiting nodes.   • Heat shock induces a systemic acquired resistance (SAR)-related gene via salicylic acid accumulation in cucumber (Cucumis sativus L.).
    Tatsuo Sato; Miyuki Kubo; Seiji Watanabe, Lead, The hypothesis that heat shock induces systemic acquired resistance (SAR) was supported by an analysis of salicylic acid (SA) content and the expression of a SAR related gene, CuPil, in cucumber. Twenty-two cultivars of cucumber were exposed to heat shock at maximum 45°C for 1 hr every day from July 7 till Sept. 14. The SA content in leaves was estimated using capillary electrophoresis. The SA content increased much more in heat-shocked leaves than in non-treated leaves in 18 varieties. However, there was no correlation between SA content and heat sensitivity in the heat tolerance test using chlorophyll fluorescence. To prove that SA induced by heat shock acts as a SAR signal transducer, the expression of CuPil, a SAR related gene, was traced using a RT-PCR-based assay. CuPil mRNA was expressed after heat shock. In conclusion, SAR appeared to be induced by heat shock without chemical inducers or infection.     佐藤達雄; 吉田 誠; 大矢武志, Lead, Pulse amplitude modulation (PAM) chlorophyll (Chl) fluorescence yield measurement was evaluated for estimating the high-temperature stress tolerance of vegetable crops. For this purpose, twenty two kinds of vegetable crops comprising of 17 taxonomic species were grown in controlled environments at the following three temperature regimes : (1) the air temperature during the 14-hr photo period was maintained at 20℃ ; (2) it was raised stepwise at 5℃ hr^<-1> from 20℃ to 35℃, and then immediately decreased stepwise to 20℃, and (3) it was changed similarly to (2) except that the duration at 35℃ was prolonged to 3 hr. At night the ambient air was cooled to 10℃ in all three pretreatments. Following the pretreatments for 48 days, photosystem (PS) II Chl. fluorescence yields of leaf sections were determined by PAM before and during exposure of leaf sections to 45℃ for up to 5 hr. Based on the heat sensitivity of PS II fluorescence yields, the vegetable crops tested were grouped into the following three classes : (A) crops with heat-tolerant PS II, (B) those with PS II sensitive to heat but capable of high-temperature acclimation, and (C) those with PS II sensitive to heat and incapable of high-temperature acclimation. These groupings coincide well with those based on the field performance of the crops at high temperatures.   • Evaluation of training and harvesting methods for vegetable jute (Corchorus olitorius L).
    Tatsuo Sato; Jiro Narimatsu, Lead, To reduce the amount of labor needed for harvesting vegetable jute, its growth habits were investigated by conventional cultivation methods. The combined marketable yields over the 50-day period were 629.1 branches/m2 and 3, 659g/m2, although a high branch density made cutting difficult in the preceding harvest. Total yield mainly consisted of the primary, secondary and third branches, whose yield peaks occurred at about 4-week intervals. The yield fluctuated at each harvest time. After harvesting the primary branches, the weights of all the other branches were about the same. Harvesting required time for the movement of scissors and time to check the branch lengths and the number of knots. To resolve these problems, a study was conducted to examine the effects of training the branches to grow in different configurations. By training the primary branches to grow horizontally and the secondary branches to grow vertically after pinching the trunk at 20cm above the ground, the marketable yield per area increased by 6% and the work time per area decreased by 17%. This method confined the cutting position mostly to a single plane and improved the efficiency of light reception.   • Wholesale harvest of vegetable jute (Corchorus olitorius L.) as a labor-saving measure.
    Tatsuo Sato; Jiro Narimatsu, Lead, To reduce the amount of labor needed to harvest vegetable jute (Corchorus olitorius L.), wholesale harvest with dense planting was examined. In this harvest method, the cutting position was fixed on a horizontal plane. Workers could use scissors at a constant height to minimize movement. As a result, by transplanting at a density of 119 plants per square meter, the marketable yield was reduced by 12%, the required time for harvest was reduced by 63%, and the marketable yield per working hour increased by 140% when compared with the values obtained by a conventional multiple harvest. By direct sowing at a density of 135 plants per square meter, the marketable yield was reduced by 28%, the required time for harvest was reduced by 87%, and the yield per working hour was increased by 442% of the values obtained by a conventional multiple harvest.
