  • Flavonoids from The Red Leaf Sheaths of Allium fistulosum ‘Hitachi-benikko’ (Ibaraki’s Specialty Red Welsh Onion Cultivar) and Their Antioxidant Activity
    Takayuki Mizuno; Risa Nakane; Takashi Kaidzuka; Ishikawa-Takano Yuko; Fumi Tatsuzawa; Eiichi Inoue and Tsukasa Iwashina, Red welsh onion (Allium fistulosum L.) ‘Hitachi-benikko’, which has red leaf sheaths, was bred by Ibaraki Agricultural Center,as a commercial vegetable in Ibaraki Prefecture. In this study, four anthocyanins including a novel glycoside [cyanidin 3-O-(3″-,O-acetyl-6″-O-malonyl)-glucoside] and five flavonols were isolated and characterized by chemical and spectroscopic methods.,The novel anthocyanin was the main component in the underground part of ‘Hitachi-benikko’. Of these flavonols, four were identified as quercetin and its glycosides, which are known to exhibit marked antioxidant activity. Furthermore, we measured the polyphenol content and antioxidant activity (H-ORAC) of the aerial and underground parts of ‘Hitachi-benikko’ and compared them with those of another general welsh onion cultivar. As the results, the reddish underground parts of ‘Hitachi-benikko’ showed higher polyphenol contents and antioxidant activities than the aerial parts of ‘Hitachi-benikko’ and those of the general,welsh onion cultivar.   • 〔Major achievements〕Transcriptome analysis of watercore in Pyrus pyrifolia by comparing pairs of susceptible and resistant F1 sibs

The disorder leads to the loss of commercial value, and resistance to watercore is one of the most important features to be studied in the breeding of Japanese pear. Even in susceptible cultivars such as 'Hosui' (syn. 'Housui'), the degree of watercore development varies markedly from year to year, which makes it difficult to analyze the characteristics of this phenomenon prior to visible watercore development. We used two F1 sib pairs from 'Hosui' x 'Hougetsu' and 'Akiakari' x 'Taihaku' crosses. Each pair contains a susceptible sib and a resistant sib. In these pairs, susceptible sibs developed severe watercore in all the fruits examined in each year, whereas no watercore was found even in overmature fruits of resistant sibs. To search for genes related to watercore, we used a custom pear microarray and studied differential gene expression in susceptible and resistant sibs prior to visible watercore development. We identified 115 differentially expressed genes including sugar metabolism-, hormone-, and cell wall-related genes. Some of these genes were differentially expressed only in immature fruits prior to watercore development. These genes would be useful as molecular markers to predict watercore at early developmental stages or in pear breeding programs., Elsevier

  • Metabolomic analyses of plasma and liver of mice fed with immature Citrus tumida peel
    Atsushi Toyoda; Mizuho Sato; Masaki Muto; Tatsuhiko Goto; Yuji Miyaguchi & Eiichi Inoue, In this study, we investigated the effects of dietary supplementation of Citrus tumida hort. ex Tanaka on food intake, body and fat tissue weights, and metabolic profiles of plasma and liver in mice. Supplementation with 5% (w/w) of peels of immature C. tumida (PIC) for 4 weeks significantly suppressed body weight gain and decreased adipose tissue weight in epididymal, perirenal, and subcutaneous fats. Metabolome analyses showed that 2-hydroxyvaleric acid levels were reduced in the blood plasma of mice fed with PIC. PIC supplementation significantly elevated dipeptide (Thr-Asp, Ser-Glu, and Ala-Ala), glucuronic acid, and S-methylglutathione levels, and significantly reduced betaine aldehyde levels in the liver. In conclusion, PIC supplementation affects the metabolism of fatty acids, pectin, glutathione, and choline, showing potential beneficial effects for metabolic syndrome and obesity.   • Dietary intake of the citrus flavonoid hesperidin affects stress-resilience and brain kynurenine levels in a subchronic and mild social defeat stress model in mice

Preventive intervention using anti-inflammatory reagents may be beneficial for alleviating the risk of depression. In this study, we focused on the Japanese local citrus plant, Citrus tumida hort. ex Tanaka (C. tumida; CT), which contains flavonoids such as hesperidin that have anti-inflammatory actions. The dietary intake of 5% immature peels of CT fruits slightly increased stress resilience in a subchronic and mild social defeat (sCSDS) model in mice. Moreover, the dietary intake of 0.1% hesperidin significantly increased stress resilience and suppressed KYN levels in the hippocampus and prefrontal cortex in these mice. In addition, KYN levels in the hippocampus and prefrontal cortex were significantly correlated with the susceptibility to stress. In conclusion, these results suggest that dietary hesperidin increases stress resilience by suppressing the augmentation of KYN signaling under sCSDS., Japan Society for Bioscience, Biotechnology, and Agrochemistry

Phylogenetic analysis of Mangifera from central region of Sumatra using trnL-F intergenic spacer. Biodiversitas 18: 1035-1040. Mangifera from Central Sumatra has an unique ability to adapt to the high rainfall regions. This genus is therefore as a potential germplasm source in Sumatra. The aim of this study was to analyze and compare the nucleotide sequences of Mangifera in the central region of Sumatra based on gene loci of trnL-F intergenic spacer. The initial step of research was conducted by isolating DNA from leaves sample of Mangifera based on CTAB method. Sequences of gene loci of trnL-F were analyzed with Maximum Parsimony and Neighbour Joining methods through PAUP* Program version 4.0b10. From the results of sequences analysis with Maximum Parsimony method, it was obtained a monophyletic group of Mangifera consisted of two main clades which separated M. kemanga from five other species of Mangifera, i.e. M. foetida, M. odorata, M. laurina and Mangifera sp. Parsimony analysis also revealed that the common ancestor of M. kemanga is the first species appeared in the central region of Sumatra. The result of Neighbour Joining analysis showed M. laurina had the longest genetic distance among examined species and might be assumed as the most primitive species of Mangifera. Mangifera laurina and M. odorata were closely related as well as M. foetida and M. odorata.   • 〔Major achievements〕De novo transcriptome assembly of mangosteen (Garcinia mangostana L.) fruit

(Mangosteen), of the family Clusiaceae, is one of the economically important tropical fruits in Indonesia. In the present study, we performed de novo transcriptomic analysis of Garcinia mangostana L. through RNA-Seq technology. We obtained the raw data from 12 libraries through Ion Proton System. Clean reads of 191,735,809 were obtained from 307,634,890 raw reads. The raw data obtained in this study can be accessible in DDBJ database with accession number of DRA005014 with bioproject accession number of PRJDB5091. We obtained 268,851 transcripts as well as 155,850 unigenes, having N50 value of 555 and 433 bp, respectively. Transcript/unigene length ranged from 201 to 5916 bp. The unigenes were annotated with two main databases from NCBI and UniProtKB, respectively having annotated-sequences of 73,287 and 73,107, respectively. These transcriptomic data will be beneficial for studying transcriptome of Garcinia mangostana L., Elsevier Inc

  • 〔Major achievements〕Analysis of Allelic Diversity and Genetic Relationships Among Cultivated Mangosteen (Garcinia mangostana L.) in Java, Indonesia Using Microsatellite Markers and Morphological Characters
    Deden Derajat Matra; Roedhy Poerwanto; Edi Santosa; Sobir; Hisao Higashio; Hiroyuki Anzai; Eiichi Inoue, The genetic variation and relationships of the mangosteen (Garcinia mangostana L.) were observed across mangosteen populations in Java, Indonesia using newly identified microsatellite loci and morphological characters. In this study, we developed an improved protocol to isolate microsatellite loci, named Selective Repeats from AFLP Sequence, by using a hybridized membrane. Twenty microsatellite loci were evaluated using 78 individuals from five mangosteen populations, and we successfully amplified four closely related Garcinia species, including G. malaccensis, G. hombroniana, G. celebica, and G. porrecta. Eight loci were monomorphic and the others were polymorphic. Sixty-nine alleles were found, with 3.491 per locus on an average. Genetic diversity (HE1) was calculated with an average across loci within population (HES1) as 0.39, an average loci across many populations (HET1) as 0.444, and genetic differentiation (FEST1) as 0.147. Furthermore, based on morphological characters, mangosteen individuals from four populations including Leuwiliang, Wanayasa, Puspahiang, and Kaligesing also had morphologically distinct fruit weight, rind weight, and rind thickness among populations. The study also elucidated the dispersal pattern of mangosteen in Java; the source of the genotype mangosteen in Java population was the Wanayasa population. In addition, we found evidence of tetraploidy in mangosteen. These results have potential applications in future breeding, conservation studies, and genetic assessment of mangosteen and their closely related species., SPRINGER

  • Effect of Yacon Tuber (Smallanthus sonchifolius)-derived Fructooligosaccharides on the Intestinal Flora and Immune System of OVA-sensitized BALB/c Mice

FOS are known as prebiotics that beneficially affect the host, i.e., improve the intestinal flora balance and enhance the immunomodulatory potential. In this study, we investigated the effect of a yacon extract rich in FOS (YOS) on the intestinal flora and immune system of BALB/c mice sensitized and challenged with ovalbumin in comparison with an extract with acid-degraded FOS (YAH). During the 4-week test period, the amount of YOS intake was lower than that of YAH; however, a significant increase in body weight was not shown among any of the test groups. Sneezing events were observed in OVA-treated mice. It was found that administration of yacon FOS improved the intestinal flora. Further, it was suggested that administration of yacon-derived FOS could epidemiologically improve the Th1/Th2 cytokine balance through its prebiotic effect on the number of beneficial bacteria in the animal, KARGER

  • Development of an Efficient Selection Method for the Parthenocarpic Trait of Tomato

In spring and autumn when self-fertilization occurs, working hours of stigma excision were shorter than,those of emasculation. The decisions of the parthenocarpic trait in each treatment generally corresponded. These findings show,that stigma excision is effective in actual breeding. In winter when self-fertilization is difficult, it is important to control night,temperature to the lowest of 8°C or higher and select individuals with seedless fruits enlarging normally from ones flowering,after the start of night temperature control.     Toshiya Yamamoto; Shingo Terakami; Norio Takada; Sogo Nishio; Noriyuki Onoue; Chikako Nishitani; Miyuki Kunihisa; Eiichi Inoue; Hiroyoshi Iwata; Takeshi Hayashi; Akihiro Itai; Toshihiro Saito, Using an F-1 population from a cross between Japanese pear (Pyrus pyrifolia Nakai) cultivars 'Akiakari' and 'Taihaku', we performed quantitative trait locus (QTL) analysis of seven fruit traits (harvest time, fruit skin color, flesh firmness, fruit weight, acid content, total soluble solids content, and preharvest fruit drop). The constructed simple sequence repeat-based genetic linkage map of 'Akiakarr consisted of 208 loci and spanned 799 cM; that of 'Taihaku. consisted of 275 loci and spanned 1039 cM. Out of significant QTLs, two QTLs for harvest time, one for fruit skin color, and one for flesh firmness were stably detected in two successive years. The QTLs for harvest time were located at the bottom of linkage group (LG) Tai3 (nearest marker: BGA35) and at the top of LG Tail5 (nearest markers: PPACS2 and MEST050), in good accordance with results of genome-wide association study. The PPACS2 gene, a member of the ACC synthase gene family, may control harvest time, preharvest fruit drop, and fruit storage potential. One major QTL associated with fruit skin color was identified at the top of LG 8.   • Evaluation of Self-fertility among Chestnut Varieties on Shading Treatment of the Flower and Genetic Analysis of the SSR Loci in Self-pollinated Seedlings
    Eiichi Inoue; Takashi Homma; Michiyasu Sasaki; Takeru Gonai; and Masakazu Kasumi, Lead, クリの自家不結実性は,他の果樹のようには完全でないと報告されているが,その自家結実性の程度については明らかになっていない。そこで本論文では,花粉遮断試験とSSR遺伝子座の遺伝解析によって,品種間における自家結実性の変異を明らかにした。花粉遮断試験の結果,調査した51品種・系統の約47%にあたる24種類において結実が確認された。そのうちの5品種とその自家結実実生13個体について,SSR遺伝子座の遺伝を確認したところ'伊吹','大峰'および'豊多摩早生'由来の8個体が自殖実生であると推察された。特に,'豊多摩早生'に関しては着毬率(62.5%)と結果率(33.3%)も他の品種・系統と比較して極めて高く,調査した6個体すべてが自殖実生であることが確認された。一方,'笠原早生','丹沢'および'大峰'に由来する5個体は,他家受粉に由来すると考えられた。以上の結果より,クリにおいては品種によっては自家結実が可能であるが,それはごく低頻度であり,基本的には自家不結実であると結論した。しかし,今回用いた品種・系統のうち,豊多摩早生'だけは自家結実性と判断した。, 一般社団法人 園芸学会

Furthermore, the effects of peeling and grating on FOS content in the root were investigated. The following results were obtained: 1) Yacon FOS decreased markedly by storage at 4 degrees C for 1 month, 2) FOS decreased by heating at 100 degrees C after peeling and grating, 3) FOS in species, except for Sarada-Otome, was little influenced by treatment with 1% acetic acid for 1. hour, 4) FOS dramatically decreased with deep-frying and was little influenced by heating by methods such as boiling, microwaving, baking and steaming, and 5) Yacon extracts from Sarada-Otome containing 4% FOS had decreased emulsifying activity and increased emulsion stability. These findings suggest that high concentrations of FOS from yacon root could be obtained by using suitable cooking conditions., JAPAN SOC FOOD SCIENCE TECHNOLOGY

  • Evaluation of polyphenol content and other properties of 16 cultivars/strains of Yacon

The tuberous roots and tubers of 3 cultivars and 13 strains of yacon were examined. Among the 3 cultivars, 'Saradaotome' and 'Saradaokame' showed superior polyphenol content in the tuberous roots. Among the 16 cultivars/strains. 'Peru-1', 'Peru-3' and 'Peru-4' showed superior polyphenol content, especially in the tuber, and 'CIP205029' showed the highest saccharide level. A positive correlation was observed between polyphenol contents in the tuberous root and tuber of the same yacon cultivar/strain.   • Development of Simple Sequence Repeat Markers in Chinese Chestnut and Their Characterization in Diverse Chestnut Cultivars
    Eiichi Inoue; Lin Ning; Hiromichi Hara; Shuan Ruan; Hiroyuki Anzai, To develop and characterize valid simple sequence repeat (SSR) markers in chestnut (Castanea spp.), we used a selective hybridization method to perform SSR sequence enrichment in the genomic library of chinese chestnut (Castanea mollissima). From 47 sequences of the enriched library, we designed 24 primer pairs for SSR polymerase chain reaction (PCR). SSR PCR was performed for 23 chinese chestnut cultivars. Among the 24 primers, 22 primers amplified their respective SSR loci; 21 primer pairs yielded polymorphic fragments across the cultivars. Among these 21 primer pairs, 17 primer pairs amplified single SSR loci with polymorphisms. Multilocus amplification patterns were observed in the other four primers. For the corresponding 17 SSR loci, the number of alleles per locus was two to 13, and the average number of alleles was 7.19. The observed heterozygosity (number of genotypes of the heterozygote/total number of genotypes scored at that locus) ranged from 0.00 to 0.87 (average = 0.68), and the expected heterozygosity ranged from 0.00 to 0.87 (average = 0.68). One of the SSR loci-ICMA014-was a unique locus, because the primer pair for this locus did not amplify any fragment in any of the cultivars, except in Qianchi, which was used to construct the SSR-enriched genomic library. Cross-species amplifications using the 17 primer pairs were also successful in the other species of genus Castanea. A dendrogram depicting the relationships among the genotypes on the basis of genetic distances was generated using the unweighted pair group method with arithmetic mean cluster analysis.   • Scanning Electron Microscopy of Strage Reserves Mobiization in Japanese Chestnut Fruit (Castanea crenata Siebold & Zucc.) during Strage Period

We stored chestnuts at −20, 2, 8 and 20°C to examine the effects of temperature on mobilization of reserves during 12-week period after harvest. At harvest (shedding stage), the cotyledonary parenchyma cells were filled with amyloplast (plastid containing a amount of substantial starch granules) and it's amount was gradually increased from the center region (adaxial region) of the cotyledon to the outer region (abaxial region) of the cotyledon. As the storage period progressed, the number of amyloplast per cell was declined in fruit stored at 2, 8 and 20°C and the rate of decline differed at each storage temperatures. Namely, higher temperatures facilitated starch decomposition. This is thought to occur because the storage temperature affects the respiration rate and starch decomposition products provide substrates for respiration. In nuts stored at −20°C, mobilization of reserves was not recognized during the storage period. The rate of starch decomposition also differed among regions of the cotyledon. At the outer region of cotyledon, the rate of decomposition was high and the inner region was low. Corresponding to the high rate of starch decomposition, an abundance of cytoplasm was observed between and surface of the amyloplast. It is suggested that the amount of cytoplasm was related to starch decomposition activities.

  • Characteristics of Interspecific Hybrids between Japanese Chestnut (Castanea crenata Sieb et Zucc.) and Seguin's Chestnut (C. seguinii Dode)
    Masakazu Kasumi; Takeru Gonai; Eiichi Inoue; Toru Manabe; Fumio Sakuma, ニホングリ(Castanea crenata Sieb. et Zucc.)とモーパングリ(C. seguinii Dode)の種間交配を行い、得られた種間雑種の特性を調査した。種間交配ではニホングリの品種交配に比べて種子形成率がやや低下するものの、発芽率はほぼ同等であった。交配実生は、RAPDによるDNA分析、形態特性から交配ミスによるニホングリ間の交配組合せと判明した個体を除くと、三度栗型実生とわい性型実生の二つに分類でき、その出現頻度はほぼ1:1に分離した。三度栗型実生は樹高がニホングリ間の交配実生と同じかやや低く、三度栗と同じように新梢に雌花を断続的に着生するものであった。わい性型実生は樹高がニホングリ間の交配実生の半分程度以下で、枝の伸長がやや少なく、結実樹齢に達するのが遅く、果実はモーパングリよりやや大きい程度(2〜3g)できわめて小さかった。このため、モーパングリを用いてわい性(3m以下)のニホングリ経済品種を育成するには、わい性を維持しつつ、‘石鎚’程度の果重(25g以上)、早期結実性(5年生までに)が必須となり、今後ニホングリとの数回の戻し交配が必要と考えられる。

  • CLONING OF A CDNA ENCODING THE SUCROSE: SUCROSE 1-FRUCTOSYLTRANSFERASE (1-SST) FROM YACON AND ITS EXPRESSION IN TRANSGENIC RICE

Anzai, Yacon is a perennial plant grown in the Andes, which was introduced into Japan for its high accumulation of inulin-type fructan in tuberous roots in 1980s. We isolated a 1-SST gene (Ye 1-SST) from cDNA library derived from yacon mature tuberous root. The coding region of the gene is 1920bp long, and its deduced protein consists of 640 amino acids with a predicted molecular mass of 72kDa. The deduced amino acid sequence of Yc 1-SST shares 87% identity with 1-SST (EC 2.4.1.99) from Jerusalem artichoke (Helianthus tuberosus L). A functional analysis for Yc 1-SST was performed by transforming into rice (Oryza saliva L. cv.Nipponbare) and compared with rice transformed with 1-SST gene of Jerusalem artichoke (Ja 1-SST) under the regulation of the cauliflower mosaic virus (CaMV) 35S promoter Transgenic rice overexpressing Yc 1-SST produced fructan such as 1-kestose (GF(2)), 1-nystose (GF(3)) and 1-fructosylnystose (GF(4)), which showed the same activity as Ja 1-SST., DIAGNOSIS PRESS LTD

  • Simple Sequence Repeat (SSR) Analysis of 7 Chestnut Cultivars in the Korean Peninsula Using SSR Markers of Japanese Chestnut
    Eiichi Inoue; Lin Ning; Toshiya Yamamoto; Shuan Ruan; Yumi Matsuki; Hiroyuki Anzai; Hiromichi Hara, Lead, Using 7 pairs of simple sequence repeat (SSR) primers developed for Japanese chestnut, we identified the SSR genotypes of 7 chestnut cultivars that originated from the Korean peninsula and compared them with those of 17 Chinese chestnut cultivars, 32 Japanese chestnut cultivars, and an interspecific hybrid obtained by crossing the above species. The 7 SSR loci of all cultivars were successfully genotyped, except KT006a for which no amplification products were obtained from 3 cultivars of Chinese chestnut. Among the 36 alleles observed in the 17 Chinese cultivars, 26 were shown to be species-specific alleles, while 37 of the 47 alleles observed in the 32 Japanese cultivars were shown to be specific to Japanese chestnut. The SSR genotypes of the Korean cultivars were also identified and comprised 2-9 alleles per locus (average: 5.14 alleles). By comparing the results to the species-specific alleles, we considered that 'Myeonsan 9', 'Myeonsan 13', 'Myeonsan 60', 'Inhungwanglyul', and 'Han 6' are Japanese chestnut cultivars; 'Hamjong 3 ho' is a Chinese chestnut cultivar; and 'Ooshiro' is an interspecific hybrid. A synonymous relationship or a bud mutation was estimated between 'Han 6' and 'Tanzawa', and between 'Ooshiro' and 'Riheiguri'.

  • Changes in Fruit Quality Factors in Japanese Chestnut (Castanea crenata Siebold & Zucc.) During Long-term Storage
    Takashi Homma; Eiichi Inoue; Toshiaki Matsuda and Hiromichi Hara, Long-term storage of Japanese chestnut (Castanea crenata Siebold & Zucc.) fruit using a polyethylene bag was carried out between October 2004 to September 2005. In order to create different O2/CO2 compositions in the bags, we used 1, 2, or 5 layers of polyethylene bags. The concentrations of soluble sugars and starch and the microstructure of cotyledonary parenchyma cells were investigated as fruit quality factors. The rate of sound fruits was decreased for all treatments at 2 months of storage. When 5 layers of polyethylene bag were used, all fruits lost their commercial quality at 10 months of storage. At 3 months of storage, there were large differences in gas composition among the treatments. The higher number of polyethylene films facilitated a decrease in the O2 content and increase in the CO2 content. Scanning electron microscopic observation of cotyledonary parenchyma cells demonstrated that the degree of starch breakdown in each treatment was also different at 3 months of storage. Starch breakdown was extensive when 1 or 2 layers of polyethylene bag were used but was decreased when 5 layers of polyethylene bag were used. These morphological differences indicate that gas composition in the bags affects fruit respiration and reserves mobilization. The amyloplast density and starch concentration in sound fruits were relatively high at 11 months of storage when 1 and 2 layers of polyethylene bag were used. At this stage, although the sound fruits rate was low (approx. 50%), a high amount of storage reserves was retained in these fruits. These findings suggest that the amount of storage reserves in chestnut fruit at harvest affects fruit storability; in other words, well-matured fruits have a good potential for long-term storage.

  • Isolation and characterization of microsatellite markers in Japanese pear (Pyrus pyrifolia Nakai)

They were characterized using 17 Japanese pear cultivars. The expected heterozygosity and observed heterozygosity ranged from 0.21 to 0.74 and from 0 to 0.88, respectively. Two to 11 alleles were detected per locus, with IPPN09 and IPPN15 judged to amplify multiple loci. IPPN17 was the most informative locus with the lowest probability of identity (0.19).   • Overcoming hybrid lethality in a cross between Japanese pear and apple using gamma irradiation and confirmation of hybrid status using flow cytometry and SSR markers

Shoots from immature hybrid embryos were gamma irradiated and cultured at normal temperature conditions to obtain viable intergencric plants. Gamma irradiation doses of 50 and 100 Gy cultured at 25 degrees C killed all shoots but there was one survivor at 150 Gy and 25 T. Hybrid status of the shoot was confirmed by using morphology, flow cytometry and SSR markers. The intergeneric plant had similar leaf serrations to apple and similar petioles to Japanese pear. Flow cytometric analysis revealed that the intergeneric shoot had a DNA content intermediate between the parents. SSR analysis showed one allele from each parent for all 16 SSR loci investigated.   • Identification of RAPD marker linked to fruit skin color in Japanese pear (Pyrus pyrifolia Nakai)

This report describes the development of a randomly amplified polymorphic DNA (RAPD) marker linked to major genes controlling the fruit skin color in Japanese pear, Two F, progenies from the cross of 'Kousui' x 'Kinchaku' and 'Niitaka' x 'Chikusui' segregated by fruit skin color were used for bulked segregant analysis. Four kinds of bulked DNA were constructed and used for the polymerase chain reaction in RAPD analysis. After 200 random primers were screened against four bulks, a band named OPH-19(425) was selected in cooperation with green bulks. The recombination rate between OPH-19(425) and the green skin phenotype was 7.3%. The RAPD marker (OPH-19425) could select green fruit with probability as high as approximately 92%. The marker was apparently useful for the selection of green-skinned individuals in a breeding program.   • Production and characterization of recombinant human lactoferrin in transgenic Javanica rice

Lactoferrin is an 80-kDa iron-binding glycoprotein that has been considered to play many biological roles, including the regulation of iron absorption, protection against microbial and virus infection, stimulation of the immune system and cellular growth promotion. We introduced two different constructs containing either the native signal peptide from human lactoferrin (pIG211) or the signal peptide from rice glutelin (pIG200) fused to mature human lactoferrin into Javanica rice cv. Rojolele by using the Agrobacterium-mediated transformation system. The expression of the hLF gene under the control of the maize ubiquitin-1 promoter was detected in all the tissues of the transgenic plants. We found that the transgenic rice plants IG200R produced a considerable amount of recombinant hLF (rhLF) in seeds, accounting for approximately 15% of the total soluble protein (TSP). RhLF was purified from mature seeds by cation-exchange chromatography. Amino acid sequencing confirmed that the N-terminal sequences of rhLF for both constructs were identical with those of native LF from human milk. Rice rhLF showed a slightly smaller molecular weight than the native hLF. Immunofluorescence microscopy revealed that rhLF was located in the intracellular and intercellular regions of endosperm cells. Protein extracts from transgenic rice seeds exhibited an antibacterial activity against Bacillus subtilis ATCC6633.   • Interspecific Hybridization of Wild Gladiolus Species Using a Dendrogram Based on RAPD Analysis

However matured seed was not obtained from the crosses using Gladiolus orchidiflorus as a female parent. We have made a segregation population (F(2)) derived from interspecific hybridization between G tristis and G gracilis for selection of a RAPD marker linked to a floral scent trait of the wild species G gracilis. Floral scent of each plant was evaluated by sensory evaluation in the F(2) plants and it was shown that a segregation ratio of scented plant and non-scented plant fit a 1:3 (chi(2-)value=0.09) ratio., International Society for Horticultural Science

  • Maternal haploidization on Japanese pear through intergeneric hybridization with apple

In the first method, we tried to induce haploidization through an inter-generic hybridization with irradiated pollen of apple. Fourteen seedlings germinated from 155 F-1 seeds that were sown. Five seedlings at 120Gy total dosage (20Gy/h dose rate) and a seedling at 80Gy (10Gy/h) survived and grew vigorously. Their morphology and DNA profiling indicated that they were similar to Japanese pear. Secondly, we tried to obtain apomicts through an inter-generic cross between a specific cultivar of Japanese pear 'Gold Nijisseiki' and apple with the intact pollen. Only one seedling survived from the 53 inter-generic seedlings that germinated. Its characteristics were also similar to the morphology of Japanese pear.   • Agrobacterium-mediated transformation of Javanica rice cv. Rojolele

Efficient callus induction from immature and mature seeds of Rojolele did not succeed by any previous method for any rice cultivar. In this study, the callus from mature seeds of Rojolele exhibited a compact and nodular appearance on C medium after the carbon source and medium pH was modified. Scutellum-derived calli from mature seeds were co-cultivated with Agrobacterium tumefaciens strains EHA101 or LBA4404 that carried plasmid pAFT14, which contained the genes for beta-glucuronidase (gus) and hygromycin resistance (hpt). Finally, the transformation efficiency of Rojolele variety using A. tumefaciens strain EHA101 (pAFT14) was improved to about 23%, similar to that of the Japonica rice variety Nipponbare. The seed fertility of transgenic Rojolele was more than 90%. The copy number of the transgene varied from one to three copies in the To transgenic lines.   • Origin and RAPD Analysis of Japanese Chestnut (Castanea crenata Sieb. et Zucc.) Cultivated in China

Though almost all the produce from Japanese chestnut trees grown in China are shipped to Japan for processing, their characterization has not been reported. In this paper, we report the origin and discrimination of varieties in China using randomly amplified polymorphic DNA (RAPD) analysis. All varieties examined in this report could be discriminated by 162 kinds of RAPD markers raised from 8 primers. Four varieties introduced to China from Japan seemed to be seedlings of the same varieties being cultivated in Japan. One of the varieties introduced from Japan, 'Moriwase' had a low similarity to 'Moriwase' in Japan. This finding suggests that 'Moriwase' in China was misinterpreted during the process by which had been introduced from Japan, or propagated via seedling. Five varieties bred in China were classified in the same group as the old varieties in Japan.

  • Effect of high-temperature on suppression of the lethality exhibited in the intergeneric hybrid between Japanese pear (Pyrus pyrifolia Nakai) and apple (Malus x domestica Borkh.)

High-temperature has been shown to be effective in suppression of hybrid lethality in rice, tobacco and cotton. If a similar response to temperature is observed in the hybrid between Japanese pear and apple, suppression of the lethality should lead to vigorous growth of the hybrid. In this experiment, we observed whether the expression of the lethality was suppressed at high-temperature or not. Clear differences in vegetative growth were observed among the shoots derived from the immature embryo at different temperature conditions. The shoots at 34°C grew vigorously without exhibiting the lethal symptoms. Similar phenomena was also observed in F1 seedlings of the cross. More than 80% of the intergeneric seedlings at high-temperature conditions (34/30°C, day/night and 34°C) grew without lethal symptoms within 18 days after sowing. Although the high-temperature suppressed the lethal symptoms, the seedlings died within 5 months from the resulting stress. In this study, we clarified that the hybrid lethality between Japanese pear and apple was suppressed at high-temperature in a similar manner to those of hybrids from wide crosses in other plant species.   • Production of apomict-like seedlings through intergeneric cross between Japanese pear and apple by means of pollen irradiation

In this experiment we tried to produce apomictic seedlings in Japanese pear through an intergeneric cross between Japanese pear and apple by means of pollen irradiation. The false hybrid can be efficiently selected in this intergeneric crossing. Because the true intergeneric hybrid potent genetic lethal system, only the false hybrid with the possibility of apomixis should survive in the crossing. Japanese pear (Pyrus pyrifolia Nakai) 'Kousui' and apple (Malus pumila Mill.) 'Fuji' were used for the intergeneric cross. The apple pollen was irradiated 6 by gamma ray from Co-60 and pollinated on the Japanese pear. A total of fourteen intergeneric seedlings at 10Gy/h (50 and 80Gy) and 20Gy/h (60 and 120Gy) developed from 155 seeds, and an individual at 80Gy (10Gy/h) and five individuals at 120Gy (20Gy/h) survived without exhibiting lethal symptoms. Plant type and morphological characters of five apomict-like seedlings at 120Gy were similar to those of Japanese pear. Their RAPD markers were only inherited from Japanese pear and their DNA contents estimated by FCM were also similar to those of Japanese pear. From the above results, we conclude that five apomict-like seedlings may be true apomicts in Japanese pear., International Society for Horticultural Science

  • Plant regeneration through leaf culture of Yacon
    Masaru Niwa,Takeomi Arai,Kazushi Fujita,Wataru Marubashi,Eiichi Inoue,Teruo Tsukihashi, Corresponding, Yacon (Smallanthus sonchifolius), a member of Asteraceae, is an ethnic food of Andeans. Interest in this crop is increasing because of its high fructo-oligosaccharide eontent in the tuberous roots. Due to the low productivity of viable seeds and virus infections through vegetative propagation, effective methods of in vitro propagation of yacon is being sought. To accomplish this, leaf pieces of yacon were cultured on the gelrite-solidified MS media, supplemented with various combinations of auxins and cytokinins. The most effective means of inducing calli was achieved on a MS medium that was supplemented with 0.1 mg・liter^<-1> 2, 4-D and 0.1 mg・liter^<-1> BAP. Shoot regeneration occurred when the calli were subcultured on the HaR medium (Paterson and Everett, 1985), supplemented with 1.0 mg・liter^<-1> BAP and 0.1 mg・liter^<-1> GA_3. By sub culturing those calli at a monthly interval, the shoot regeneration continued up to the 10th subculture. The regenerated shoots rooted ; the plantlets were well-acclimatized and transplanted successfully to the field. AFLP analysis conducted on the extracted DNA from the regenerants revealed 12 out of the 13 possessed different amplified fragments from those of the mother plants, indicating that the somaclonal mutation took place during the course of callusing and plant regeneration., 一般社団法人 園芸学会

  • Evaluation of germplasm collection in wild Gladiolus species of Southern africa
    Yasumasa Takatsu; Toru Manabe; Masakazu Kasumi; Tetsuya Yamada; Ryuji Aoki; Eiichi Inoue; Yoich Morinaka; Wataru Marubashi; Mikio Hayashi, 21種のグラジオラス野生種について特性調査および育種素材としての評価を行ったところ, 草丈, 葉数, 葉·花の形態, 小花数および開花期等には種によって大きな違いがみられた. また草丈が10cm程度で鉢物に利用可能なもの, 現在の栽培種にはみられない青色の花被片を有するものなど, 育種素材として有望な野生種が見出された. 香りを有する種は全体の52.3%を占め, 香りのタイプもチョウジ様, スミレ様などさまざまであることが示された. さらにこれらの野生種について到花日数, 小花の開花期間, 稔実日数および1さや当たりの種子数を調査し育種上重要な情報を得ることができた. フローサイトメトリーによる解析の結果, 野生種においては細胞あたりのDNA含量が多様で, 種によってイネの0.9∼3.5倍のゲノムサイズを有するものと推定された. 本法による倍数性の判定は困難であるが, 種の組合せによっては交雑後代の雑種性の検定に利用可能であることが示唆された., 日本育種学会

  • Interspecific hybridization of wild gladiolus species of southern Africa, based on random amplified polymorphic DNA markers

Out of the 140 tested primers, 32 amplified a total of 133 RAPD bands in 33 Gladiolus species. The genetic distance was calculated from the data of these RAPDs, and a dendrogram was generated. Interspecific crosses were carried out in seven combinations within or between clusters, and F1 seedlings were obtained from most combinations. The RAPD analysis showed that these F1 seedlings were real hybrids. The results suggest that RAPD markers are useful for detecting genetic relationships in Gladiolus species, and for interspecific crosses in breeding programs. (C) 2001 Elsevier Science B.V.   • Tempareture effects on interspecific hybridization between Gladiolus x grandiflora and G. tristis

(2n = 60) and the wild species G. tristis L. (2n = 30) was made to introduce characteristics of the wild species into the cultivated one. Gladiolus xgrandiflora is a summer-flowering species, and G. tristis flowers in winter. The effect of storage temperature on pollen viability was tested, as long-term storage of pollen was necessary to facilitate crossing these two species. Pollen of G. tristis could he stored st -20 degreesC for approximate to1 year, and was more practical than storage at -80 degreesC. Air temperature affected pollen tube growth, fertility, and fruit set in the cross between G. xgrandiflora and G. tristis, and low temperatures (15 to 20 degreesC) were best. The morphological data and flow cytometric analysis showed that the F(1) plants were hybrids between G. xgrandiflora and G. tristis., American Society for Horticultural Science

  • Characterization and overcoming of temperature dependent lethality exhibited by the hybrid embryos from the cross Nicotiana suaveolens x N. sylvestris

The immature embryos of the cross Nicotiana suaveolens x N. sylvestris did not grow and could not be rescued by conventional ovule culture. We observed that culture of the ovules at high temperatures (30, 32, 34 degreesC) enabled to suppress this obstacle temporarily. Although many plantlets were obtained through ovule culture at high temperatures, the plants did not grow to maturity. On the other hand, ovule culture with a high concentration of cytokinin was effective in overcoming the obstacle. In ovule culture on 1/2 MS + BAP (10 mg/l), many hybrid plantlets of the cross N. suaveolens x N. sylvestris developed and formed vigorous multiple shoots. The hybrid plants obtained from multiple shoots grew vigorously and matured in a greenhouse. Because the characteristics of the obstacle were similar to those of hybrid lethality exhibited by the seedlings of the cross between N. suaveolus and N. tabacum, we suggested that the obstacle was due to hybrid lethality that developed just after fertilization. Furthermore, we were able to obtain many mature hybrid plantlets in the cross N. suaveolens x N. sylvestris by ovule culture on a medium containing cytokinin.   • Intergeneric crossing between Japanese pear (Pyrus pyrifolia Nakai) and Apple (Malus pumila Mill.)
    Fumio Sakuma; Eiichi Inoue; Masakazu Kasumi, ニホンナシにリンゴ果実の持つ芳香性や高糖度,貯蔵性など優良な形質を導入しようとして,ニホンナシとリンゴの属間で正逆交雑を試みた。その結果,ニホンナシを種子親とした場合,4613花交配して21.7%の結果率であった。一方リンゴを種子親にした場合は,2232花交配して9.3%の結果率であった。特に,ニホンナシ'幸水'およびリンゴ'ふじ'を種子親にした交配組み合わせで結果率が高かった。しかし,ニホンナシを種子親にした場合22.1%,リンゴを種子親にした場合11.1%の無種子果が発生した。特に,ニホンナシ'幸水'を種子親,リンゴ'王林'を種子親または花粉親とした場合無種子果の発生が多かった。属間交配果の稔実種子数は,ニホンナシを種子親とした場合は1果当たり2.0個で品種間交配の2.7個より少なかった。また,リンゴを種子親にした場合は1果当たり1.2個で品種間交配の2.3個より減少した。しかし,種子重に大きな差はみられなかった。属間交雑種子の発芽率は,ニホンナシを種子親を種子親にした場合1013粒播種して61.6%,リンゴを種子親にした場合117粒播種して29.1%であった。それに対して品種間交雑種子の発芽率は,ニホンナシでは59.5%,リンゴでは81.3%でリンゴを種子親にした属間交配で発芽率がかなり劣った。実生は播種16日後から5ヶ月の間に雑種致死のためすべて枯死した。交配70日後の胚を培養することによりシュートが得られた。胚培養由来シュートは34℃の高温条件下で雑種致死を抑制できたが,やはり枯死し雑種致死は回避できなかった。また,胚培養により得られ,30日毎に継代培養しているシュートはWPM培地でピロガロールおよびIBA処理により発根が促進できたが,順化後枯死した。, 茨城県農業総合センター生物工学研究所

In the liquid media containing 0.01-10 mg/l cytokinin, shoot-like structures were formed at the hypocotyl and the primary root of the dying seedlings. The shoot-like structures did not exhibit lethality and grew up to vigorous shoots without developing any lethal symptoms within a month. Vigorous multiple shoots developed from the shoot-like structures in the liquid media containing more than 0.2 mg/l cytokinins. The regenerated shoots were transferred to the medium containing 2 mg/l indole-3-acetic acid (IAA) to accelerate rooting. The hybrid seedlings displayed a vigorous growth without developing any lethal symptoms up to maturity in a greenhouse. Forty chromosomes were counted in root tip cells of the seedlings and morphological characters of the seedlings were intermediate between those of parental species.   • Genomic factors controlling the lethality exhibited in the hybrid between Nicotiana suaveolens Lehm and N-tabacum L
    E Inoue; W Marubashi; M Niwa, Interspecific hybrid plants between Nicotiana suaveolens and N. tabacum exhibit lethal symptoms at the seedling stage and cannot grow to maturity. In this investigation, an attempt was made to clarify the genomic factors responsible for this lethality. N. suaveolens was crossed to N. sylvestris (genomic constitution: SS) and N. tomentosiformis (TT), these latter two species being the progenitors of N. tabacum (SSTT). From the cross N. suaveolens x N. tomentosiformis, many seedlings were obtained through ovule culture, and these subsequently grew to maturity without exhibiting any lethality. In the reciprocal crossing between N. sauvelons and N. sylvestris, only a few hybrid seedlings were obtained through ovlue culture and all died after unfolding their cotyledons when cultured at 28 degrees C. This lethality could be avoided by culturing the ovules at 36 degrees C. These features of hybrid lethality resembled those observed in the interspecific hybrid between N. suaveolens and N. tabacum. These findings suggest that the S genome in N. tabacum is responsible for the lethality exhibited in the hybrid between N. suaveolens and N. tabacum., SPRINGER VERLAG
