Yasue T.; N.Sakamoto; N.Hirayama; M.Komatsuzaki; Y.Yamakawa; H.Takahashi; S.Shibanuma; D.Kohari; T.Okayama; A.Toyoda, 日本型の園芸療法(CF)を確立するため、統合失調症および/または器質性精神障害をもつ成人17名(男性14名、女性3名、21~64歳)を対象に検討した。農作業は、30分間の園芸作業(種まきおよび収穫)と、30分間の家畜との触れ合い(ウシとヤギのブラッシングおよび給餌)から成り、二つの作業は1ヵ月おきに入れ替えた。作業中は心拍数を連続的に記録し、心理的ストレス反応尺度(SRS)と唾液中α-アミラーゼ濃度(sAA)を各作業の前と後に測定した。心拍数変動分析では、交感神経と副交感神経の興奮の指標であるLF/HFとHF nuを作業の前と後で比較した。sAA濃度は作業の前と後で変わらなかったが、SRSスコアは抑鬱/不安の項目で有意に低下した。LF/HFとHF nuも、園芸作業後に家畜と触れ合うことにより有意に異なっていた。これらの所見から、二つを組合わせたCFは、精神疾患をもつ成人の不安や緊張を軽減する可能性があると考えられた。, ヒトと動物の関係学会

Tsuyoshi Okayama; Yoshifumi Kubota; Atsushi Toyoda; Daisuke Kohari; Go Noguchi, A noninvasive method for estimating the body weight (BW) of a pig considering its posture using a low-cost depth camera (Kinect v2) was proposed. A total of 150 pigs were used, and 738 depth images (point clouds) were obtained for them. The pig "volume" was calculated from the pig point cloud, and it was found to have a very high correlation to BW. To evaluate the posture of a pig quantitatively, seven posture angles were calculated based on the "spine" extracted from a pig point cloud. We found the posture angles representing the height of the head position correlated with the accuracy of BW estimation using the "volume." Based on this finding, we proposed an "adjusted volume," which was adjusted based on the relationship between the posture angles and the estimation error. The BW of pigs was estimated using the simple regression model with the "adjusted volume," and the MAPE and RMSPE were 4.87% and 6.13%, respectively. The accuracy of the suggested model was similar to that of the volume-based estimation models of other studies that used only data with an appropriate pig posture for BW estimation., 日本畜産学会

Herein, we performed a comprehensive analysis of the CoA pools formed in thirteen tissues of Zucker and Zucker diabetic fatty (ZDF) rats. Hypothalamic malonyl-CoA levels in obese rats remained low and were almost the same as those of lean rats, despite obese rats having much higher content of leptin, insulin, and glucose in their sera. Regardless of the fa-genotypes, larger total CoA pools were formed in the livers of ZDF rats and the size of hepatic total CoA pools in Zucker rats showed almost one tenth of the size of ZDF rats. The decreased total CoA pool sizes in Zucker rats was observed in the brown adipose tissues, while ZDF-fatty rats possessed 6% of total CoA pool in the lean rats in response to fa deficiency. This substantially lower CoA content in the obese rats would be disadvantageous to non-shivering thermogenesis. Thus, comparing the intracellular CoA behaviors between Zucker and ZDF rats, as well as the lean and fatty rats of each strain would help to elucidate features of obesity and type 2 diabetes in combination with result (s) of differential gene expression analysis and/or comparative genomics.

To develop drug therapies for the treatment of depression, it is necessary to use animal models of depression to screen drug candidates that exhibit anti-depressive effects. Unfortunately, the present methods of drug screening for antidepressants, the forced-swim test and tail-suspension test, are limiting factors in drug discovery because they are not based on the constructive validity of objective phenotypes in depression. Previously, we discovered that the onset of nest building is severely delayed in mice exposed to subchronic mild social defeat stress (sCSDS). Therefore, a novel paradigm combining acute social defeat stress (ASDS) and the nest-building test (SNB) were established for the efficient screening of drugs for depressive-like symptoms. Since ASDS severely delayed the nest-building process as shown in chronically social defeated mice, we sought to rescue the delayed nest-building behavior in ASDS mice. Injecting a specific serotonin 2a receptor antagonist (SR-46349B), the nest-building deficit exhibited by ASDS mice was partially rescued. On the other hand, a selective serotonin reuptake inhibitor (fluoxetine) did not rescue the nest building deficit in ASDS mice. Therefore, we conclude that the SNB paradigm is an another potential behavioral method for screening drugs for depressive-like symptoms including attention deficit, anxiety, low locomotion, and decreased motivation. BEHAVIOURAL PROCESSES, Feb. 2017, [Reviewed] - Behavioral restriction effects on activity motivation of a captive lion (Panthera leo persica)

However, these reasons for movement and locomotion differ among species. This study investigated the locomotive motivation of captive lions, which are regarded as having relatively low motivation to move compared to other zoo animals. Four Indian lions including 1 noncastrated male and 3 females were observed as subject animals. Two captive conditions were set as experimental days: 1 was a confined condition in a bed chamber all day long (confined day); another was a released condition, with access to the exhibition area during daytime (released day). The observation periods were also defined as "daytime" (9:00-16:00) and "nighttime" (16:30-9:00 of the next morning). We counted the lions' behavior repertories and recorded their maintenance, social, reproductive, and abnormal pacing behavior of 24 hours each day 5 times from the beginning of August to the end of November. Results show no difference in the numbers of behavior repertories between the released and confined days (chi(2)=0.084, NS), suggesting that the conditions in this zoo might be sufficient for captive lions to express various behavior repertories. However, the maintenance behavior percentages were significantly different when confined and released days were compared (daytime, chi(2) = 19.17, P < 0.01; nighttime, chi(2) = 13.06, P < 0.05). All the lions rested nearly all day long except around evening housing time or in the morning on the released days. However, on confined days, they were usually restless. Furthermore, abnormal pacing during walking (chi(2) = 3.94, P < 0.05) was significantly different on each day. These results suggest that the lion's locomotor needs were not satisfied when the lions were confined in the bed chamber all day, even though they did not always move actively. Four guide dogs were used as subjects. Asphalt, gravel, and water floors were set up as experiment road conditions. During the experiment, the trainer and a subject dog with a harness walked along these straight paths (9m×2-1.8m). A 3D accelerometer was attached to the loin of each dog, with six markers attached at the carpus, elbow, point of shoulder, hock joint, stifle, and major trochanter. Their gait was recorded from their left side using a video recorder. The symmetry, regularity, and stability of the gait rhythm were evaluated using acceleration data with fast Fourier transformation. The kinetic state of the gait was evaluated by the maximum and minimum angle of elbow and hock joints, maximum height of front and rear pads, and stride length for each road condition. The gait velocity of each dog and trainer was also measured from video images. No difference was found in their gait rhythms, maximum or minimum angles of elbow and hock joints, maximum heights of front pads, or stride lengths in these road conditions. However, the maximum height of the rear pad in gravel was higher than that on asphalt (P<0.05) or on a water floor (P=0.05). Tha gait velocity in gravel tended to be lower than that on a water floor (P=0.072). The gait velocity gap between a dog and human in gravel tended to be greater than that on a water floor (P=0.074). Results show that different road conditions can affect the gait velocity and guide walking, but not the gait rhythm or kinetic state of guide dogs., Japanese Soceity of Livestock Management

高倉 梓; 路川 強; 田中 正夫; 小針 大助, 黒毛和種育成子牛における誘導訓練が,ロープ誘導能率,対人反応性および誘導前後の安寧性に及ぼす影響について調査を行った。育成子牛7頭を用いて誘導訓練および最終試験を実施し,誘導時間,誘導反応,逃走距離および心拍反応の変化を記録した。誘導時間は訓練の経過に伴う有意な変化はなかったが,個体間で有意差が見られた(P<0.05)。変動係数を求めたところ,訓練1週目では特に個体により大きなバラつきが見られたが,2週目以降は低く安定した。抵抗反応は,訓練期間の経過に伴い減少する傾向が見られ(P=0.064),経路間では有意差が見られた(P<.0005)。また暴走反応は,最終試験で多く発生した。逃走距離および心拍反応は,いずれも有意差は見られなかった。本結果から,育成牛においても誘導訓練の実施により個体による誘導時間の不安定さや抵抗反応は減少するが,人への慣れから模擬闘争行動が増加する可能性が示唆された。, 日本家畜管理学会

A relatively larger pool size of total CoA, consisting of acetyl-CoA, malonyl-CoA, and CoASH, was observed in the medulla oblongata, liver, heart, and brown adipose tissue. Focusing on changes in the CoA pool size in response to the nutrient composition of the diet given, total CoA pools in rats continuously fed a high-fat diet for 4 weeks were significantly higher in the hypothalamus, cerebellum, and kidney, and significantly lower in the liver and skeletal muscle than those of rats fed a high-carbohydrate or high-protein diet. In particular, reductions in the liver were remarkable and were caused by decreased CoASH levels. Consequently, the total CoA pool size was reduced by approximately one-fifth of the hepatic contents of rats fed the other diets. In the hypothalamus, which monitors energy balance, all three CoA molecular species measured were at higher levels when rats were fed the high-fat diet. Thus, it was of interest that feeding rats a high-fat diet affected the behaviors of CoA pools in the hypothalamus, liver, and skeletal muscle, suggesting a significant relationship between CoA pools, especially malonyl-CoA and/or CoASH pools, and lipid metabolism in vivo. Biochemical and Biophysical Research Communications, 2012, [Reviewed] - Effects of chronic social defeat stress on MAP kinase cascade

In this study, we analyzed the mitogen-activated protein kinase (MAPK) cascade in the hippocampus of chronically socially defeated rats. The rats that were subjected to social defeat every day for 5 weeks showed physiological and behavioral changes, including a reduced rate of weight gain, enlarged adrenal glands, and increased immobility in the forced swim test without concomitant changes in locomotor activity in the open field test. Altered body weight and enlarged adrenal glands are typical symptoms of human depression. Prolonged immobility in the forced swim test indicates behavioral despair, a well-established index of depression. Because the MAPK cascade plays a pivotal role in depression, we quantified the expression of these molecules in the hippocampus of chronically defeated rats using western blot analysis. We found that phospho-MAPK kinases 1/2 (MEK1/2) and phospho-extracellular signal-regulated kinases 1/2 (ERK1/2) were decreased, whereas MAPK phosphatase-1 (MKP-1) was increased in the hippocampus of chronically defeated rats compared to the control group. These results were consistent with findings in depressed patients and other animal models of depression. In conclusion, our findings suggest that chronic psychosocial stress in Wistar rats induced depression-like behavior and downregulated the MAPK cascade in the hippocampus. Daisuke Kohari; Kazuya Akimoto; Michiru Fukasawa, The aim of this study was to clarify the relationships between flight distance (FD) and the physiological and behavioral responses in cows during handling. We measured the FDs of 16 Japanese Black cows that are not routinely handled and selected the 4 longest (LFD) and 4 shortest FD cows (SFD). During the handling test, each cow was tethered and brushed by same person who conducted the FD test. Behavioral responses (vocalization, tail swinging, head shaking and stepping) and heart rate (HR) of each cow were recorded before, during and after the handling period (5min per period). Behavioral responses and the mean HR (MHR) of each period, the percentage of time the HR during handling was higher than MHR before handling (PHR) and the recovery times from the first peaks during handling to MHR before handling (RT) were compared between the LFD and SFD cows. No difference between LFD and SFD cows was found for any behavioral response. However, the MHR (P<0.05) and PHR (P=0.05) were higher in LFD than in SFD cows. Furthermore, RT was longer in LFD than in SFD cows (P=0.07). Animal Behaviour and Management, 2010 - The synchronization of behavioural response of calves at weaning by time lag weaning in Japanese Black Cattle.
Sachiko Sugihara; Daisuke Kohari, We investigated the synchronization of behavioural responses of calves at weaning using time-lag weaning method. Six 4-month-old Japanese Black Cattle calves were used as experimental animals. They were divided into control (n=3) and delayed weaning groups (n=3). Then they weaned in pairs. During the eight-day experiment, DW calves were weaned a day after control calves. We observed vocalization and self-grooming of each calf through continuous observations. Maintenance behaviours (lying at rest, eating, and walking) and changes of location in the experimental paddock were recorded by 1-min instantaneous sampling. Vocalizations during the week following separation and self-grooming during the week and during each observation day were compared among pairs using the Mann-Whitney U-test. The paddock was divided into three blocks: front, centre, and back. The concordance rates of the staying blocks were calculated. Furthermore, the rate of movement (m/h) was measured for each pair and was used for comparison. Vocalizations of control calves were more numerous than those of the DW group (control, 3641±673 calls; DW, 3384±715 calls), but not significantly so. Furthermore, the groups' vocalization peaks differed. Self-grooming incidences were significantly more numerous in the control group than in the DW group: 291±56 times, 122±36 times, respectively. A significant difference was found during 3-6 days after weaning (P<0.05). Regarding maintenance behaviour, the transitions of eating and walking were similar in each pair. The concordance rates of the staying blocks were extremely high among experimental pairs: 95.6%, 86.6%, and 82.1%, respectively. A moving distance peak was observed in the second day the DW group (the second day of control). The transition was similar for each pair. 塚田英晴; 深澤充; 小迫孝実; 小針大助; 佐藤衆介, At a private farm where forest grazing was started in 2004, effects of cattle on the abundance of small mammals and their habitats were evaluated. Small mammals were trapped during 2003-2007; their numbers at grazing and non-grazing forest sites were compared. Four characteristics of small mammals' habitats at grazing and non-grazing forest sites, i.e. vegetation structure, plant cover, litter thickness, and soil hardness, were investigated and compared. Numbers of small mammals at grazing and nongrazing forest sites did not differ. No significant cumulative effect of 4 years' forest grazing was found. Although three habitat qualities of small mammals, vegetation structure, plant cover and litter thickness, were different between grazing and non-grazing forest sites from the non-grazing condition identified at the start of this study, these habitat differences showed no significant cumulative effect attributable to 4 years' forest grazing. Five Japanese Black breeding cows were used as experimental cattle. Three approach directions were tested; that were the approach toward the head of the test cow from the front direction (FRONT), the approach toward to the foreleg of the test cow from the right or left (SIDE) and the approach toward to hind-leg of the test cow diagonally from the right or left rear (BACK). Three levels of approach pace were also tested; that were the approach at a pace of 7 km/h, the approach at a pace of 4 km/h, and the approach at a pace of 1 km/h. The shortest distance between observer and cow's at the moment of the cow's withdrawal was defined as the flight distance. The flight distance in FRONT was significantly shorter than that in SIDE. The effect of the approach pace was significant. The faster an observer approached to cow, the greater the flight distance shown by the cow. The manner of approach to cattle markedly affects the flight distance. Therefore, the manner of approach to cattle must be standardized to measure flight distance for sharing results obtained by several researchers under various conditions., WILEY-BLACKWELL PUBLISHING, INC

Fukasawa Michiru; Tsukada Hideharu; Kosako Takami; Kohari Daisuke., The present study examined and discussed the way the basal diet (BD) during the preliminary phase and the last diet (LD) before the food selection test affected the selective feeding behavior of heifers. For the BD and LD, oat hay and concentrate were used. Three different levels of roughage content (low, medium and high) were prepared by changing the mix ratios of oat hay and concentrate. Low and high roughage diets were used for the BD, and all three of the roughage levels were used for the LD. A mixture of orchard grass and white clover (MIX), and Japanese pampas grass leaf (JP) were used as testing foods. In the preliminary phase, all heifers were fed the BD three times per day. In the test phase, heifers were fed the LD for the first meal (06.00-12.00 hours), and food selection under different BD and LD was examined during the second meal (13.00-16.00 hours) of the day. The effects of the BD were significant for MIX dry matter intake (DMI), JP DMI and the rate of MIX DMI for total DMI. Under the high roughage BD condition, heifers ingested more MIX than under the low roughage BD condition. The effect of the LD was significant on JP DMI, whereas the rate of MIX DMI for total DMI was not significant. Selective feeding behavior was strongly affected by the BD and slightly by the LD.

Daisuke Kohari; Shusuke Sato; Kazuo Sugawara., Maternal grooming (MG), nursing (NS) and mother-infant behavior 5 min before (IB5) and immediately before (LIB) maternal behavior were observed in Japanese Black (JB) and Japanese Shorthorn (JS) cow-calf pairs. During MG of the JB, the number of calf's IB5 was more than that of cows, but the duration of the MG positively correlated with the number of cow's IB5. During MG of the JS, the duration positively correlated with the number of calf's IB5. The number of the NS with calf's LIB and the number of calf's IBS in the NS were more than those of cows in both breeds. The duration of the NS tended to correlate with the number of calf's IBS in the JS. Main behavior sequence was observed like cow's approach, MG, calf's contact and NS, in this order, in both breeds. It is thought that the MG is released by cow's and calf's activities in both breeds and accelerated by the cow's activeness in the JB and by the calf's activeness in the JS, and that the NS is released and accelerated by the calf's activeness in JS.

小針大助; 佐藤衆介; 菅原和夫, Maternal behaviour of 17 multiparous cows and the growth of their calves were observed in Japanese Black cattle. Total durations of maternal grooming and nursing behaviour and the percentage of cow-calf distance within 5 m as maternal behaviour were investigated using focal animal sampling for 6 h from 30 min before sunrise at 2 nd day and 1 st, 2 nd, 3 rd, 4 th weeks after parturition. Temperament, dominance value and parity were evaluated as the dam's character. In addition, the daily weight gain (DG) and the duration of scours were investigated in each calf during the first month after a birth as the indices of growth. As the genetic factor of sires may influence the DGs of their calves, DGs of sires in the individual performance test and the progeny test were also investigated. Individual differences were observed in the total duration of maternal grooming (P<0.0001) and the percentage of cow-calf distance within 5 m (P<0.005), but not the total duration of nursing behaviour. There were no correlations among the total durations of maternal grooming and nursing behaviour and the percentage of cow-calf distance within 5 m, that suggests that 3 aspects of maternal behaviour are independent each other. The total duration of maternal grooming correlated with the calf's DG (r=0.64, P<0.005) and the day of scours (r=-0.67, P<0.05). The percentage of cow-calf distance within 5 m correlated with dominance value (r=0.54, P<0.005) and parity of a dam (r=0.74, P<0.005).
